目的明确生殖周期中卵巢甾体激素波动是否引发特定脑区神经元突触外GABAA受体表达变化及相关性。方法阴道涂片法筛选出具有完整生殖周期的大鼠,并将其分为两组,动情期组与动情间期组;Elisa检测每组大鼠血清雌孕激素含量;免疫组化法观察每组大鼠海马齿状回、杏仁核突触外GABAA受体δ亚基的表达;分析各组大鼠血清孕激素含量与突触外GABAA受体δ亚基表达的相关性。结果动情期组血清雌激素含量高于动情间期组,而孕酮含量则低于动情间期组,组间差异具有统计学意义(P<0.05)。海马齿状回、杏仁核GABAA受体δ亚基的免疫组化结果显示,阳性表达动情间期组高于动情期组,差异具有统计学意义(P<0.05);海马齿状回、杏仁核突触外δ亚基阳性结果 IOD值随着动情间期与动情期孕酮浓度值的增加而增加,两变量呈线性相关。结论动情间期高水平的孕激素可致大脑海马齿状回、杏仁核突触外GABAA受体高表达。
OBJECTIVE: To determine whether ovarian steroid hormone fluctuations in the reproductive cycle lead to changes in the expression of extra-synaptic GABAA receptors in specific brain regions and their correlation. Methods Vaginal smear method was used to screen rats with complete reproductive cycle and divided into two groups: estrus group and estrus period group; Elisa serum estradiol and progesterone content of each group; immunohistochemistry The expression of GABAA receptor δ subunit in hippocampal dentate gyrus and amygdala nucleus was detected by immunohistochemistry. The correlation between serum progesterone level and the expression of subunit of GABAA receptor δ subunit in each group was analyzed. Results The content of estrogen in estrous period group was higher than that in estrous period group, but the content of progesterone in estrous period group was lower than that in estrous period group. The difference between the two groups was statistically significant (P <0.05). The hippocampal dentate gyrus, the amygdala GABAA receptor δ subunit immunohistochemical results showed that positive expression of estrous interval group was higher than that of estrus group, the difference was statistically significant (P <0.05); hippocampal dentate gyrus, amygdala The IOD value of the positive results of the extrasynaptic delta subunit increased with the increase of estrus interval and estrus progesterone concentration, and the two variables were linearly correlated. Conclusion The high estrogen level of progesterone can induce the hippocampal dentate gyrus and the high expression of GABAA receptor outside the amygdala.
