目的 研究苦参碱对不同分化程度的胃癌细胞株在体外的杀伤作用及其机制。方法 用不同剂量的苦参碱分别作用于中、低分化胃癌细胞株SGC-7901和AGS 24、48、72h,以MTT法检测药物对肿瘤细胞的杀伤作用;透射电镜和流式细胞术检测苦参碱诱导SGC-7901凋亡的作用。结果 随药物浓度的增加和作用时间的延长,苦参碱对胃癌细胞的杀伤作用逐渐增强(P<0.01);低分化的AGS较中分化的SGC-7901更为敏感。透射电镜、流式细胞术检测均显示苦参碱能够诱导胃癌细胞凋亡。结论苦参碱在体外对胃癌细胞具有杀伤作用,且呈明显的量效和时效关系;低分化的胃癌细胞株AGS对苦参碱更为敏感;苦参碱对胃癌细胞株的体外杀伤作用与其诱导胃癌细胞凋亡有关。
Objective To study the killing effect and mechanism of matrine on gastric cancer cell lines with different differentiation in vitro. Methods Different doses of matrine were applied to SGC-7901 and AGS 24, 48, and 72 h respectively. MTT assay was used to detect the killing effect on tumor cells. Results With the increase of drug concentration and prolongation of action time, the killing effect of matrine on gastric cancer cells gradually increased (P <0.01). The poorly differentiated AGS was more sensitive than the moderately differentiated SGC-7901. Transmission electron microscopy and flow cytometry showed that matrine could induce gastric cancer cell apoptosis. Conclusion Matrine has a killing effect on gastric cancer cells in vitro and shows a significant dose-effect and time-dependent relationship. Matrine is more sensitive to matrine than matrine, and matrine can kill gastric cancer cells in vitro Induction of gastric cancer cell apoptosis.
