目的观察在人直肠癌组织中分化抑制因子-2(Id-2)和基质金属蛋白酶-9(MMP-9)的表达情况,分析两者表达水平的相关性,探讨直肠癌临床病理学指标与两者的表达水平的关系。方法收集直肠癌组织和癌旁正常组织标本56例,采用免疫组织化学方法观察Id-2和MMP-9在直肠癌和癌旁正常组织中的表达水平,Spearman相关分析法分析Id-2与MMP-9表达有无相关性,分析Id-2与MMP-9表达与直肠癌临床病理学指标之间的关系。结果 Id-2在直肠癌组织的阳性表达率明显高于癌旁正常组织(73.21%vs.48.21%,P<0.05),MMP-9在直肠癌组织中的阳性表达率高于癌旁正常组织(71.43%vs.44.64%,P<0.05)。Spearman相关分析检测Id-2与MMP-9表达水平存在正相关性(r=0.393,P=0.003);Id-2和MMP-9表达水平与直肠癌分化程度、TNM分期和淋巴结转移有关(P<0.05);而与年龄和性别无关(P>0.05)。结论 Id-2和MMP-9的表达水平与直肠癌的发生发展存在着密切的关系,直肠癌组织中Id-2高表达可能是通过MMP-9途径来促进肿瘤的侵袭转移。
Objective To observe the expression of Id-2 and MMP-9 in human rectal cancer and to analyze the correlation between the expression of Id-2 and MMP-9 and the clinicopathological parameters of rectal cancer The relationship between the two expression levels. Methods 56 cases of colorectal cancer tissues and adjacent normal tissue specimens were collected. The expression of Id-2 and MMP-9 in rectal cancer tissues and adjacent normal tissues was observed by immunohistochemistry. Spearman correlation analysis was used to analyze the correlation between Id-2 and MMP -9 expression or not, the relationship between Id-2 and MMP-9 expression and clinicopathological parameters of rectal cancer were analyzed. Results The positive expression rate of Id-2 in rectal cancer tissues was significantly higher than that in adjacent normal tissues (73.21% vs.48.21%, P <0.05). The positive expression rate of MMP-9 in rectal cancer tissues was higher than that in adjacent normal tissues (71.43% vs.44.64%, P <0.05). There was a positive correlation between the expression of Id-2 and MMP-9 by Spearman correlation analysis (r = 0.393, P = 0.003). The expression of Id-2 and MMP-9 was correlated with the differentiation degree, TNM stage and lymph node metastasis <0.05), but not related to age and gender (P> 0.05). Conclusion The expression of Id-2 and MMP-9 is closely related to the occurrence and development of rectal cancer. The high expression of Id-2 in rectal cancer may be through the MMP-9 pathway to promote tumor invasion and metastasis.
