使用数字相机配合计算机组合成一套设备“计算机数字图像分析系统(GDAS)”,分析测定分子生物学研究中SDS-PAGE凝胶电泳所分离的目标蛋白相对分子质量。提出了利用凝胶图像的像素及蛋白区带的质心来确定其迁移率。并根据标准蛋白的迁移率拟合曲线,通过测量目标蛋白的迁移率,利用拟合曲线方程计算出目标蛋白的相对分子质量。测量结果表明,利用质心确定蛋白区带的迁移率,所得到的结果,与理论值比较误差较小。
Using a digital camera combined with a computer, a set of equipment, “Computer Numerical Image Analysis System (GDAS)”, was used to analyze and determine the relative molecular mass of the target protein isolated by SDS-PAGE gel electrophoresis in molecular biology research. It is proposed to use the centroid of the pixel and protein bands of the gel image to determine the mobility. The curve of the standard protein mobility was fitted and the relative molecular mass of the target protein was calculated by using the fitted curve equation by measuring the mobility of the target protein. The measurement results show that using the centroid to determine the mobility of the protein band, the obtained result is less error compared with the theoretical value.
