目的研究下丘脑和海马内c-fos和c-jun蛋白的表达与应激反应之间的时效关系。方法采用特异性抗体的原位免疫细胞化学方法,对游泳应激后不同时间点的大鼠下丘脑和海马内fos和jun蛋白阳性神经元的分布进行观察,用图像分析技术对大鼠下丘脑室旁核(PVN)、视上核(SON)和海马齿状回(DG)内的fos和jun阳性细胞的相对切面面积比和平均目标灰度进行分析。结果游泳应激结束后3h,室旁核和视上校及齿状区内可见密集深染的c-fos和c-jun阳性细胞,室旁核小细胞部,背内侧核等区域亦有疏密不一的阳性胞体。图像分析结果发现,游泳应激结束后3~8h内下丘脑PVN和SON及海马DG内fos和jun阳性胞体的相对切面面积比明显高于正常大鼠(P<0.01),而平均目标灰度则显著降低(P<0.01)。结论应激可以诱导下丘脑和海马等相关脑区c-fos和c-jun的表达,并且具有很强的时效关系。
Objective To study the relationship between the expression of c-fos and c-jun protein in hypothalamus and hippocampus and the stress response. Methods In situ immunocytochemistry was used to observe the distribution of fos and jun protein positive neurons in the hypothalamus and hippocampus of rats at different time points after swimming stress. By using image analysis technique, the hypothalamus Paraventricular nucleus (PVN), supraoptic nucleus (SON) and hippocampal dentate gyrus (DG) in the fos and jun positive cells relative to the area ratio and the average target gray-scale analysis. Results The c-fos and c-jun positive cells were observed in paraventricular nucleus, paraventricular nucleus and dentate zone 3h after the end of swimming stress. There were also some small areas in the paraventricular nucleus and dorsal medial nucleus Different positive cell body. The results of image analysis showed that the relative cross-sectional area ratio of fos and jun positive soma of PVN and SON in hypothalamus and hippocampus DG in 3 ~ 8 h after swimming stress were significantly higher than that in normal rats (P <0.01) Was significantly lower (P <0.01). Conclusion Stress can induce the expression of c-fos and c-jun in the relevant brain regions such as the hypothalamus and hippocampus, and have a strong time-dependent relationship.
