目的建立合欢皮水溶性木脂素类化合物的纯化工艺。方法以合欢皮木脂素苷H为指标,首先对大孔树脂种类进行筛选,然后对选定的大孔树脂进行纯化工艺参数优化。结果 D101型大孔树脂对合欢皮水溶性木脂素类纯化效果最佳,最佳工艺为:上样液的浓度为0.4g·ml-1,上样流速为2BV·h-1,树脂径高比为1:6,上样量为1.8倍树脂量,先用5BV的水以5VB·h-1流速洗脱除杂,再用40%的乙醇溶液作为洗脱剂以2BV·h-1流速洗脱,洗脱体积为5BV。结论该方法简单易行,分离效果好。
OBJECTIVE To establish a purification process of water-soluble lignan compounds of Acacia bark. Methods Hesperidin H as an index, first of all macroporous resin species were screened, and then the selected macroporous resin purification process parameters optimization. Results The results showed that D101 macroporous resin had the best purification effect on water-soluble lignans of Acacia flavescens. The optimum conditions were as follows: the concentration of sample solution was 0.4g · ml-1, the flow rate of sample was 2BV · h-1, The ratio of 1: 6, the amount of sample 1.8 times the amount of resin, first with 5BV water to 5VB · h-1 flow rate impurity removal, and then 40% ethanol solution as eluent to 2BV · h-1 Flow rate elution, elution volume of 5BV. Conclusion The method is simple and easy to operate with good separation effect.
