目的观察乳鼠心肌细胞移植对兔急性心肌梗死区微血管密度及微血管再生的影响。方法新西兰大白兔16只,新生乳鼠40只。建立兔急性心肌梗死模型后,将培养的SD乳鼠心肌细胞移植到兔心肌梗死区,分别于移植后1、23、、4周取材,用碱性磷酸酶组织化学法和免疫荧光法进行染色,光镜观察,LEICA Qwin V3图像分析系统分析兔心肌梗死边缘区毛细血管密度及其再生情况。结果乳鼠心肌细胞移植后的1、2、3、4周不同的时间段,兔心肌梗死区的毛细血管密度及新生毛细血管数量均明显高于对照组(P<0.05)。定量分析显示,毛细血管密度呈现先升后降的趋势。结论异体异种心肌细胞移植能够促进兔心肌梗死区的毛细血管新生,有利于改善心肌的血液供应。
Objective To observe the effect of neonatal rat cardiomyocyte transplantation on the microvessel density and microvascular regeneration in acute myocardial infarction area in rabbits. Methods 16 New Zealand white rabbits and 40 newborn rats. After establishment of a rabbit model of acute myocardial infarction, cultured neonatal rat cardiomyocytes were transplanted into rabbit myocardial infarction area and were harvested at 1, 23, 4 weeks after transplantation, and stained with alkaline phosphatase histochemistry and immunofluorescence , Light microscopy, LEICA Qwin V3 image analysis system analysis of myocardial infarction in rabbit capillary density and regeneration. Results At 1, 2, 3, 4 weeks after myocardial cell transplantation, the capillary density and the number of capillaries in myocardial infarction area were significantly higher than those in control group (P <0.05). Quantitative analysis showed that the capillary density showed a trend of first rising and then decreasing. Conclusion Allogeneic cardiomyocyte transplantation can promote capillary angiogenesis in rabbit myocardial infarction area, which is beneficial to improve myocardial blood supply.
