目前对肥大细胞研究的焦点之一就是肥大细胞的异质性。本实验对比观察了大鼠皮肤结缔组织肥大细胞(CTMC)和胃肠粘膜肥大细胞(MMC)在甲苯胺兰染色性和细胞内肽类胺类物质免疫反应性方面的异同。结果表明:经甲醛固定的皮肤CTMC用甲苯胺兰常规染色即可显示,面臂肠MMC需低pH并延长染色时间才能显示;结果可见几乎全部皮肤CTMC免疫组织化学染色,显示5—羟色胺免疫反应性,仅10%的CTMC显示P物质免疫反应性:胃肠MMC的35%显示5-羟色胺免疫反应性,未见P物质免疫反应性。结果提示肥大细胞异质性不仅表现在甲苯胺兰染色,而且也表现在免疫组织化学染色上。
One of the current focuses on mast cells is the mast cell heterogeneity. In this experiment, the similarities and differences between tolmatamine staining and intracellular peptide amines immunoreactivity were observed in rat skin connective tissue mast cells (CTMC) and gastrointestinal mucosa mast cells (MMC). The results showed that formalin-fixed skin CTMC with toluidine blue staining can be shown, facial arm intestinal MMC need low pH and prolonged staining time can be displayed; results can be seen almost all skin CTMC immunohistochemical staining showed serotonin immune response Only 10% of CTMC showed substance P immunoreactivity: 35% of gastrointestinal MMC showed serotonin immunoreactivity and no substance P immunoreactivity was seen. The results suggest that mast cell heterogeneity is not only demonstrated by toluidine blue staining, but also by immunohistochemical staining.
