目的:研究骨髓增生异常综合征(MDS)患者骨髓中CD34(+)细胞自噬活性,并探讨其临床意义。方法:收集2012年10月至2014年3月我院收治的20例MDS、20例非恶性病贫血和5例急性髓系白血病(AML)患者骨髓液单个核细胞(BMMNC),采用流式细胞术检测这些标本中CD34(+)细胞自噬活性。结果:低危组MDS及非恶性病贫血组CD34(+)细胞自噬活性高于高危组MDS及AML组(P<0.05),且MDS患者中CD34(+)细胞自噬活性与WHO分型预后积分系统(WPSS)呈负相关(r=-0.887)。结论:MDS患者CD34(+)细胞自噬活性较AM L患者明显增高,同时M DS患者骨髓CD34(+)细胞自噬活性与M DS患者WPSS评分呈负相关,提示自噬活性的降低有可能促进MDS疾病的演进,同时与MDS的预后相关。
Objective: To study the autophagy activity of CD34 (+) cells in bone marrow of patients with myelodysplastic syndrome (MDS) and to investigate its clinical significance. Methods: Bone marrow mononuclear cells (BMMNC) from 20 patients with MDS, 20 patients with non-malignant anemia and 5 patients with acute myeloid leukemia (AML) were collected from October 2012 to March 2014 in our hospital. Flow cytometry The autophagy activity of CD34 (+) cells in these specimens was examined. Results: The autophagic activity of CD34 (+) cells in MDS and non-malignant anemia groups was higher than that in high-risk MDS and AML groups (P <0.05), and the autophagic activity of CD34 (+ Prognostic Score System (WPSS) was negatively correlated (r = -0.887). CONCLUSIONS: The autophagy activity of CD34 (+) cells in patients with MDS is significantly higher than that in patients with AML, while the autophagy activity of CD34 (+) cells in M DS patients is negatively correlated with WPSS score in patients with M DS, suggesting a possible reduction of autophagy activity Promotes the progression of MDS disease, and is associated with the prognosis of MDS.
