目的:探讨黏附分子E-钙黏蛋白(E-cadherin)的表达对人滋养细胞侵袭能力的调控及其与自然流产发生的关系。方法:分别以过表达或小干扰RNA(siRNA)的方法上调或下调人绒毛膜癌细胞株Bewo及JEG-3内源性E-钙黏蛋白表达,Transwell侵袭实验观察细胞侵袭行为的改变,并应用Western blotting比较4例正常妊娠妇女及4例自然流产妇女绒毛组织内E-钙黏蛋白的表达水平。结果:转染野生型E-钙黏蛋白质粒的Bewo及JEG-3细胞随着E-钙黏蛋白表达的提高,细胞的侵袭能力显著下降(P<0.05);而转染E-钙黏蛋白siRNA的Bewo及JEG-3细胞,随着E-钙黏蛋白表达的下降,细胞侵袭能力显著提高(P<0.05)。自然流产妇女较正常妊娠妇女的绒毛组织E-钙黏蛋白的表达水平不同程度出现升高。结论:E-钙黏蛋白参与调控滋养细胞的侵袭能力,其表达水平可能与妊娠结局密切相关。
Objective: To investigate the regulation of human trophoblast invasion and its relation with spontaneous abortion by the expression of E-cadherin. Methods: The expression of endogenous E-cadherin in human choriocarcinoma cell lines Bewo and JEG-3 was up-regulated or down regulated by overexpression or small interfering RNA (siRNA) respectively. Transwell invasion assay was used to observe the change of cell invasive behavior The expression of E-cadherin in villi of 4 normal pregnant women and 4 spontaneous abortion women were compared by Western blotting. Results: The invasion ability of Bewo and JEG-3 cells transfected with wild-type E-cadherin decreased with the increase of E-cadherin (P <0.05), while the transfection of E-cadherin siRNA Bewo and JEG-3 cells, with the decline of E-cadherin expression, cell invasion significantly increased (P <0.05). Natural abortion women than normal pregnant women, the villus tissue E-cadherin expression levels increased to varying degrees. Conclusion: E-cadherin is involved in the regulation of trophoblast invasion and its expression level may be closely related to pregnancy outcome.
