目的研究恶性髓系肿瘤细胞中部分岩藻糖转移酶(FUT1-3、FUT6-7)基因的相对表达情况。方法应用实时荧光定量逆转录-聚合酶链反应(RT-PCR)检测HL-60细胞及31名急性髓细胞白血病(AML)患者与骨髓增生异常综合征(MDS)患者(患者组)的FUT1-3、FUT6-7基因的相对表达情况,同时以正常CD34+细胞作为HL-60细胞的正常对照、以25名健康体检者作为患者组的正常对照。结果 FUT1-3、FUT6-7基因在HL-60细胞及其对照组的相对表达分别为:0.30±0.03 vs 1.08±0.06、0.68±0.05 vs 1.20±0.14、0.68±0.04 vs 1.00±0.05、0.82±0.03 vs 1.00±0.05、0.59±0.12 vs 1.02±0.15(P<0.01)。FUT1、FUT3基因在患者组与对照组的相对表达分别为:0.85±0.67 vs 1.31±0.5、0.53±0.48 vs 1.34±0.69(P<0.05);FUT2、FUT6、FUT7基因的相对表达量在患者组与对照组间无明显差异(P>0.05)。FUT1-3、FUT6-7基因在AML患者(n=13)与MDS患者(n=18)间的相对表达量差异亦不明显(P>0.05)。结论 FUT1-3、FUT6-7基因在恶性髓系肿瘤细胞HL-60及AML与MDS患者中出现不同程度的相对表达减弱,二者间部分具有关联性。
Objective To study the relative expression of some fucosyltransferases (FUT1-3, FUT6-7) in malignant myeloid tumor cells. Methods Real-time fluorescent quantitative reverse transcription-polymerase chain reaction (RT-PCR) was used to detect the expression of FUT1-β in HL-60 cells and 31 patients with acute myeloid leukemia (AML) and myelodysplastic syndrome (MDS) The normal expression of FUT6-7 gene, normal CD34 + cells as a normal control HL-60 cells, 25 healthy subjects as the normal control group. Results The relative expression of FUT1-3 and FUT6-7 in HL-60 cells and their control groups were: 0.30 ± 0.03 vs 1.08 ± 0.06, 0.68 ± 0.05 vs 1.20 ± 0.14, 0.68 ± 0.04 vs 1.00 ± 0.05, 0.82 ± 0.03 vs 1.00 ± 0.05, 0.59 ± 0.12 vs 1.02 ± 0.15 (P <0.01). The relative expression of FUT1, FUT3 and FUT3 in patients and control groups were 0.85 ± 0.67 vs 1.31 ± 0.5,0.53 ± 0.48 vs 1.34 ± 0.69 (P <0.05) No significant difference with the control group (P> 0.05). The relative expression levels of FUT1-3 and FUT6-7 genes between AML patients (n = 13) and MDS patients (n = 18) were also not significantly different (P> 0.05). Conclusions The expression of FUT1-3 and FUT6-7 in malignant myeloid tumor cells HL-60 and AML and MDS patients are weakened to some extent, and some of them are related to each other.
