消黄方对α-萘异硫氰酸酯大鼠黄疸模型肝组织氧化应激的影响

目的:探讨消黄方对α-萘异硫氰酸酯(ANIT)大鼠黄疸模型的治疗作用及抗氧化应激效应。方法:Wistar雄性大鼠40只,随机分为正常组(10只)和ANIT处理组(30只)。ANIT按照2mL/kg一次性经口灌胃,48h后,ANIT处理组随机分为模型组,消黄方组及熊去氧胆酸(UDCA)组(各10只),各组每天分别给予生理盐水、消黄方、UDCA治疗1周,实验结束处死全部大鼠。观测大鼠死亡率、检测肝功能及肝组织总超氧化物歧化酶(T-SOD)、谷胱甘肽-S转移酶(GST)活性和丙二醛(MDA)含量。结果:肝功能结果发现:与正常组比较,模型组大鼠肝ALT、AST、ALP、TBIL、DBIL显著升高(P<0.05,P<0.01)。与模型组比较,UDCA和消黄方显著降低血清TBIL和DBIL含量(P<0.01),消黄方显著降低ALT、AST及ALP活性(P<0.05,P<0.01)。肝组织氧化应激结果显示:与正常组比较,模型组肝组织GST活性及MDA含量显著升高(P<0.01),T-SOD活性显著下降(P<0.01),而消黄方显著降低GST活性及MDA含量(P<0.01),升高肝组织SOD活性(P<0.01)。结论:消黄方具有保肝退黄作用;氧化应激是ANIT大鼠黄疸模型的重要病理环节,抗氧化应激效应是消黄方退黄主要机制之一。 Objective: To investigate the therapeutic effect of Xiaohuang Recipe on jaundice in rats induced by α-naphthalene isothiocyanate (ANIT) and its anti-oxidative stress effects. Methods: Forty Wistar male rats were randomly divided into normal group (n = 10) and ANIT group (n = 30). ANIT was administered orally once a day at 2 mL / kg. After 48 h, ANIT-treated rats were randomly divided into model group, Xiahuang Fang group and UDCA group (10 rats in each) Saline, Xiaohuang Fang, UDCA treatment for 1 week, all rats were sacrificed at the end of the experiment. The mortality of rats was observed. The liver function and the content of T-SOD, GST and the content of malondialdehyde (MDA) in the liver were detected. Results: The results of liver function test showed that compared with the normal group, ALT, AST, ALP, TBIL and DBIL in the model group were significantly increased (P <0.05, P <0.01). Compared with the model group, UDCA and Xiaoxian prescription significantly decreased the levels of serum TBIL and DBIL (P <0.01), Xiaoya recipe significantly reduced the activity of ALT, AST and ALP (P <0.05, P <0.01). The results of oxidative stress in liver tissue showed that the activity of GST and the content of MDA in model group were significantly increased (P <0.01) and the activity of T-SOD was significantly decreased (P <0.01) compared with normal group Activity and MDA content (P <0.01), and increased the activity of SOD in liver tissue (P <0.01). Conclusion: Xia Huang Fang has the function of protecting the liver from yellowing. Oxidative stress is an important pathological link in the model of jaundice in ANIT rats. Antioxidant stress is one of the main mechanisms of Xia Huang decay yellow.
