①目的对低温保存有活性生物瓣膜和血管的效果优劣进行了酶组织化学的检测,并观察消毒液对组织的损伤程度。②方法标本取自新鲜整体猪,分为新鲜对照、24h灭菌后不同保护剂冻贮、未经24h灭菌不同保护剂冻贮、经碘扑溶液消毒后的新鲜和冻贮组织等共计7组。均进行琥珀酸脱氢酶、核苷酸酶、磷酸化酶、葡萄糖-6-磷酸脱氢酶和乳酸脱氢酶的组织化学半定量检测。③结果标本在低温保存前先行24h灭菌对组织无明显不利的影响;实验组经低温保存后其5种酶活性与新鲜对照组相近,但低温保护剂为M199组稍好于HSA组;将组织浸泡于1%碘扑溶液中2min,对酶活性有轻度影响。④结论低温保存活性血管和瓣膜以M199+DMSO组效果最佳;使用碘扑溶液行心内组织消毒时应尽量缩短其作用时间
①Objective To detect the effect of preservation of active biological valves and blood vessels at low temperature by enzyme histochemistry and observe the degree of tissue damage caused by disinfectant. ② Methods The specimens were collected from fresh whole pigs and divided into fresh control, frozen and stored with different protective agents after 24h sterilization, frozen and stored with different protective agents without 24h sterilization, fresh and frozen tissues after disinfection by iodophor solution, etc. Total 7 group. Histochemical semi-quantitative detection of succinate dehydrogenase, nucleotidase, phosphorylase, glucose-6-phosphate dehydrogenase and lactate dehydrogenase were performed. (3) The results showed that there was no significant adverse effect on the tissue sterilized before sterilized in low temperature. The activity of the five enzymes in the experimental group after cryopreservation was similar to that in the fresh control group, but the hypothermia protective agent was slightly better in the M199 group than the HSA group. Tissue soaked in 1% iodophor solution for 2min, a slight impact on enzyme activity. ④ Conclusion Cryopreservation of activated blood vessels and valves to M199 + DMSO group best; use of iodophor solution of intra-ventricular tissue should be disinfected to minimize the role of time
