[目的]研究长链非编码RNAs(lncRNAs)在刀豆蛋白A(ConA)诱导的小鼠免疫性肝损伤肝组织中表达谱的变化。[方法]利用lncRNAs芯片技术分别检测正常对照组小鼠及ConA诱导的免疫性肝损伤小鼠肝组织中lncRNAs表达谱,经对原始数据进行预处理达到均一化后,筛选出差异表达的lncRNAs并进行分析。[结果]与正常对照组小鼠相比较,ConA诱导的免疫性肝损伤在小鼠肝组织中表达1.5倍以上并有显著差异(P<0.05)的lncRNAs,确定为差异表达的lncRNAs。1.5倍以上变化的共60条;1.5倍以上升高的共17条;1.5倍以上降低的共43条;2倍以上升高共4条;2倍以上降低的共8条;3倍以上升高共2条;3倍以上降低的共2条。[结论]与正常对照组小鼠肝组织比较,ConA诱导的免疫性肝损伤小鼠肝组织中lncRNAs表达谱发生显著变化,提示差异性表达的lncRNAs可能参与了ConA诱导的小鼠免疫性肝损伤发生。
[Objective] To investigate the changes of expression profiles of long-chain non-coding RNAs (lncRNAs) in liver tissue induced by ConA in mice with immunological liver injury. [Methods] lncRNAs were used to detect the expression profiles of lncRNAs in the liver of normal control mice and ConA-induced immunological liver injury mice respectively. After the original data were preprocessed to be uniform, the differentially expressed lncRNAs were screened Analyze. [Results] Compared with normal control mice, lncRNAs expressed more than 1.5-fold in the liver tissue of mice with ConA-induced immune injury were significantly different (P <0.05), and were identified as differentially expressed lncRNAs. 1.5 times more than a total of 60 changes; 1.5 times more than a total of 17; 1.5 times more than a total of 43; 2 times more than a total of 4; 2 times more than a total of 8; 3 times ascending A total of 2 high; 3 times lower than the total of 2. [Conclusion] The expression of lncRNAs in liver tissues of mice with immune-induced liver injury induced by ConA changed significantly compared with that of the normal control mice, suggesting that differentially expressed lncRNAs may be involved in ConA-induced immune liver injury in mice occur.
