目的:研究癫痫清颗粒对β-淀粉样蛋白(Aβ_(25-35))致阿尔茨海默病模型(AD)小鼠的学习记忆改善作用。方法:60只昆明种鼠随机分为假手术组,模型组,癫痫清颗粒低(3.12 g·kg~(-1))、中(6.24 g·kg~(-1))和高(9.36 g·kg~(-1))剂量组和盐酸多奈哌齐组(1.30 mg·kg~(-1)),海马内注射(Aβ_(25-35)),制作小鼠AD模型。连续灌胃给药15 d。Y迷宫和Morris水迷宫实验检测各组小鼠认知行为改变,HE染色观察其对AD模型小鼠神经元的影响。结果:癫痫清颗粒可显著增加AD模型小鼠的自发交替反应率,缩短逃避潜伏期,减少总路程,延长游泳路程百分比,可改善海马区神经元细胞排列紊乱、水肿等现象,减少神经元死亡。结论:癫痫清颗粒对Aβ_(25-35)致阿尔茨海默病模型小鼠学习记忆能力具有改善作用。
Objective: To study the effect of epilepsy-clearing granule on learning and memory of Alzheimer’s disease (AD) mice induced by β-amyloid (Aβ_ (25-35)). Methods: Sixty Kunming mice were randomly divided into sham operation group, model group, epilepsy clearance granules (3.12 g · kg -1), medium (6.24 g · kg -1) and high (9.36 g Kg -1) and donepezil hydrochloride (1.30 mg · kg -1) and Aβ 25-25 (injection of hippocampus). Continuous oral administration of 15 d. Y maze and Morris water maze test to detect the cognitive behavior of mice in each group, HE staining was used to observe the effect of AD model mice neurons. Results: Epilepsia Granules can significantly increase the spontaneous alternation response rate, shorten the escape latency, decrease the total distance and prolong the swimming distance in AD model mice, and can improve the disorganization and edema of neurons in hippocampus and reduce the neuronal death. Conclusion: Epilepsi Capsule can improve the learning and memory abilities of mice with Alzheimer’s disease induced by Aβ 25-35.
