采用FISH-AFLP技术,选取8对EcoRⅠ+3和MseⅠ+3引物组合,对新疆维吾尔自治区林木品种审定委员会审(认)定的24个核桃品种(无性系)、2株实生古树及4个农家类型在DNA水平上进行了多态性检测。结果表明:在获得的1 011条谱带中,981条呈多态性,多态性百分率达97.5%;不同引物组合的有效等位基因数为1.188 71.234 7,平均为1.208 5;基因多样度为0.118 3 0.141 2,平均为0.129 7;Shannon信息指数为0.184 6 0.225 8,平均为0.206 6。总体的遗传多样性水平为中等。经8对引物检测的30个品种及类型均得到数目不等的特异带型,能将30个核桃良种及类型完全区分,建立了它们的指纹图谱。
Eight pairs of EcoRⅠ + 3 and MseⅠ + 3 primer combinations were selected by FISH-AFLP technique. Twenty-four walnut cultivars (clones), two provenance trees and four Farm type Polymorphism was detected at the DNA level. The results showed that among the 1 011 bands obtained, 981 were polymorphic and the percentage of polymorphism was 97.5%. The effective number of alleles in different primer combinations was 1.188 71.234 7 with an average of 1.208 5. The genetic diversity 0.118 3 0.141 2 with an average of 0.129 7 and Shannon Information Index 0.184 6 0.225 8 with an average of 0.206 6. The overall level of genetic diversity is medium. The 30 varieties and types tested by 8 pairs of primers all got special bands with different numbers. Thirty kinds of walnut varieties and types were completely distinguished, and their fingerprints were established.
