目的 探讨细胞周期调控抑制因子p5 7kip2 蛋白和cyclinE(周期蛋白E)在大肠腺癌发生发展中的作用。方法 采用免疫组化技术SP法 ,检测p5 7kip2 和cyclinE蛋白在 3 7例大肠腺癌和 2 6例正常大肠粘膜组织中的表达。结果 p5 7kip2 蛋白阳性表达率在大肠腺癌组织中为 40 5 % ,显著低于正常大肠粘膜组织 ( χ2 =5 0 3 9,P <0 0 5 ) ,并与大肠腺癌组织分化程度和淋巴结转移均有关 ( χ2 =8 914 ,χ2 =4 416,P <0 0 5 ) ;cyclinE蛋白阳性表达率在大肠腺癌组织中为 64 9% ,显著高于正常大肠粘膜组织 ( χ2 =4 2 85 ,P <0 0 5 ) ,并与大肠腺癌组织分化程度和淋巴结转移均有关 ( χ2 =8 3 97,χ2 =5 2 61,P <0 0 5 ) ;p5 7kip2 蛋白阳性表达组cyclinE蛋白阳性表达率低于p5 7kip2 蛋白阴性表达组 ,但两者之间呈负相关 ( r=-0 3 0 2 7,P <0 0 5 )。结论 p5 7kip2 蛋白的低表达和 (或 )cyclinE蛋白的高表达可能在大肠腺癌发生发展中发挥重要作用 ;p5 7kip2 和cyclinE蛋白在大肠腺癌细胞周期调控中可能存在着负反馈调节机制
Objective To investigate the role of cell cycle regulatory inhibitor p5 7kip2 and cyclin E in the development of colorectal adenocarcinoma. Methods Immunohistochemical SP method was used to detect the expression of p5 7kip2 and cyclinE protein in 37 cases of colorectal adenocarcinoma and 26 cases of normal colorectal mucosa. Results The positive rate of p5 7kip2 protein in colorectal adenocarcinoma was 40.5%, which was significantly lower than that in normal colorectal mucosa (χ2 = 5093, P <0 05) (Χ2 = 8 914, χ2 = 4 416, P <0 05). The positive expression rate of cyclinE protein in colon adenocarcinoma was 64.9%, which was significantly higher than that in normal colorectal mucosa (χ2 = 4285 , P <0.05), and were correlated with histological grade and lymph node metastasis of colorectal adenocarcinoma (χ2 = 8 3 97, χ2 = 5261, P <0 05). The positive expression of cyclinE protein in p5 7kip2 positive group The expression rate was lower than p5 7kip2 negative expression group, but negative correlation was found between them (r = -0.0327, P <0.05). Conclusions The low expression of p5 7kip2 protein and / or overexpression of cyclinE protein may play an important role in the development of colorectal adenocarcinoma. There may be negative feedback regulation mechanism of p5 7kip2 and cyclinE protein in the cell cycle of colorectal adenocarcinoma
