目的:建立胰腺癌裸鼠皮下及原位模型的方法,并为胰腺癌体内实验提供实验基础。方法:利用人胰腺癌细胞SW1990制备细胞悬液给予皮下荷瘤鼠组腋下种植,记录原位组肿瘤长径短径病计算肿瘤体积,并测量体重,原位组为将细胞悬液原位移植于裸鼠胰腺,从而建立裸鼠原位种植胰腺癌模型。结果:各组均未出现术后死亡情况,术后成瘤率为100%。接种后两组接种后体重均变化平稳,皮下移植瘤组肿瘤体积于接种第五天后呈指数增长,第九天达到125.6±20.02mm3,原位模型组则可于体表触及凸起肿块。结论:建立了两种稳定可靠成瘤率高的胰腺癌动物模型,为胰腺癌的研究提供了基础。
Objective: To establish a subcutaneous and in situ model of pancreatic cancer in nude mice and to provide experimental basis for in vivo experiments of pancreatic cancer. Methods: The human pancreatic cancer cell line SW1990 was used to prepare the cell suspension for subcutaneous tumor-bearing mice. The tumor volume was calculated and the body weight was measured. The in situ group was the cell suspension in situ Transplanted into the pancreas of nude mice to establish a model of pancreatic cancer in situ implantation in nude mice. Results: No postoperative death occurred in all groups. The rate of tumorigenesis after operation was 100%. After inoculation, the weights of the two groups changed smoothly. The tumor volume of subcutaneous xenograft group increased exponentially on the fifth day after inoculation, reached 125.6 ± 20.02mm3 on the ninth day, while the in situ model group could reach the protruding mass on the body surface. Conclusion: Two kinds of animal models of pancreatic cancer with stable and reliable tumor formation rate were established, which provided the basis for the study of pancreatic cancer.
