采用胶原酶的单酶冷消化法,分离培养37例肾细胞癌患者的肿瘤浸润淋巴细胞(tumor infiltra-ting lymphocytes,TILs)。结果发现,采用该法分离培养的 TILs 呈现以激活的 T 淋巴细胞为主、高增殖活性和高杀伤活性的特征。逆转录-多聚酶链式反应(RT-PCR)检测结果提示,TILs 激活扩增后主要分泌产生肿瘤坏死因子-α(TNF-α),其可能在 TILs 发挥抗肿瘤活性中起重要作用。应用结果显示,治疗后患者外周血淋巴细胞表型分析,90%以上患者 CD3~+细胞和70%患者 CD8~+细胞得到提高。机体的细胞免疫水平提高。
Tumor infiltrating lymphocytes (TILs) were isolated and cultured in 37 patients with renal cell carcinoma by single enzyme digestion with collagenase. The results showed that the TILs isolated and cultured by this method showed the characteristics of activated T lymphocytes, high proliferative activity and high killing activity. The result of reverse transcription-polymerase chain reaction (RT-PCR) suggested that TILs secreted mainly produce TNF-α, which may play an important role in the antitumor activity of TILs. Application results show that after treatment, peripheral blood lymphocyte phenotype analysis in patients, more than 90% of patients with CD3 ~ + cells and 70% of patients with CD8 ~ + cells increased. The body’s cellular immunity is increased.
