目的:研究五味温通除痹胶囊对佐剂性关节炎大鼠细胞因子的调控作用。方法:雄性SD大鼠60只,随机分成6组,每组10只。除正常对照组外,采用弗氏完全佐剂诱导AA大鼠模型。造模第12天灌胃给予相应药物,1次/d,连续12 d。实验结束后,酶联免疫吸附法测定血清IL-1、IL-4、IL-6、IL-10及TNF-α的含量;同时取固定部位踝关节组织,HE染色观察病理学改变;RT-PCR技术测定滑膜组织中IL-1、TNF-αmRNA的表达。结果:与模型组比较,五味温通除痹胶囊(1.60、3.20 g/kg)不仅能减轻大鼠关节病理损伤程度,还能明显抑制IL-1、IL-6、TNF-α的表达,升高IL-4、IL-10的水平。结论:五味温通除痹胶囊对AA大鼠具有一定的治疗作用,其机制可能与抑制促炎因子并促进抑炎因子的产生,调控细胞因子网络平衡有关。
OBJECTIVE: To study the regulation effect of Wutong Wentongbi Capsule on cytokines in adjuvant arthritis rats. Methods: Sixty male Sprague-Dawley rats were randomly divided into 6 groups with 10 rats in each group. In addition to the normal control group, complete Freund’s adjuvant induced AA rat model. On the 12th day, the rats were given gavage with the corresponding drugs once a day for 12 days. At the end of the experiment, the levels of IL-1, IL-4, IL-6, IL-10 and TNF-αwere measured by enzyme linked immunosorbent assay. PCR technique was used to detect the expression of IL-1 and TNF-αmRNA in synovial tissue. Results: Compared with the model group, Wuwei Wensheng Tongbi Capsule (1.60,3.20 g / kg) can not only reduce the degree of pathological injury in rats, but also inhibit the expression of IL-1, IL-6 and TNF- High IL-4, IL-10 levels. CONCLUSION: Wuwei Weitong BiBu Capsule has a certain therapeutic effect on AA rats. The mechanism may be related to the inhibition of proinflammatory cytokines and the production of anti-inflammatory cytokines and the regulation of cytokine network balance.
