目的从神经-内分泌-免疫角度来探讨慢性疲劳综合征的发生机制及龟鹿益神颗粒的调节作用。方法采用慢性束缚和夹尾激怒加强制冷水游泳方法,建立复合应激致慢性疲劳大鼠模型。造模成功后,模型组、龟鹿益神颗粒组、苁蓉益肾颗粒组分别给予生理盐水、龟鹿益神颗粒混悬液、苁蓉益肾颗粒混悬液灌胃治疗,共计14 d。用ELISA法测定各组大鼠血浆β-EP和血清IL-2含量。结果造模后各组大鼠体重增长缓慢甚至减轻。龟鹿益神颗粒、苁蓉益肾颗粒组IL-2含量升高、β-EP含量降低,且龟鹿益神颗粒组IL-2含量高于苁蓉益肾颗粒组、β-EP含量低于苁蓉益肾颗粒组。结论龟鹿益神颗粒可通过调整机体内阿片肽(β-EP)及IL-2的含量,对抗应激,改善机体免疫功能而对神经-内分泌-免疫网络调节产生影响,从而改善应激所致的疲劳。
Objective To explore the mechanism of chronic fatigue syndrome and the regulating role of Gui Lu Yi Shen Granule from the perspective of neuroendocrine-immunology. Methods Chronic restraint and tail-wrapping anger were used to enhance the cooling water swimming method, and a rat model of chronic fatigue induced by combined stress was established. After successful modeling, the model group, Guigu Lu Yishen granule group, Cistanche Yishen granule group were given saline, Guigu de Yishen granule suspension, Cistanche Yishen granule suspension gavage treatment, a total of 14 d . The levels of plasma β-EP and serum IL-2 in each group were determined by ELISA. Results After modeling, the weight gain of rats in each group increased slowly or even reduced. The content of IL-2 in Cistanche Yishen Granule group and the content ofβ-EP in Guixue Yishen Granule group were higher than those in Cistanche Yishen Granule group, and the content of β-EP was low Cistanche Yishen granule group. Conclusion Guiguyizhen granule can affect neuro-endocrine-immune network regulation by adjusting the content of opioid peptide (β-EP) and IL-2 in the body, counteracting stress and improving immune function, Caused by fatigue.
