目的:探讨不同转移能力的肺癌细胞株中miRNA表达差异,并分析miRNA在肺癌转移过程中的功能及意义。方法:利用非编码小RNA芯片技术,检测高转移肺癌细胞株(PGBE1)与低转移肺癌细胞株(PGLH7)间miRNA的表达差异,并通过qPCR鉴定。结果:非编码小RNA芯片分析显示,在不同转移能力肺癌细胞中得到4条差异表达的miRNA,分别为hsa-mir-34b、hsa-mir-23a、hsa-mir-23b和hsa-mir-199a-5p。实时定量PCR对差异的miRNA进行鉴定,4条差异的miRNA在低转移细胞株(PGLH7)中的丰度均高于其在高转移细胞株(PGBE1)中的丰度,P<0.05,与芯片结果吻合。结论:4条miRNA可能与肺癌转移相关,对miRNA功能探讨将有助于研究肺癌转移的分子机制。
Objective: To investigate the miRNA expression in lung cancer cell lines with different metastatic potential and to analyze the function and significance of miRNA in lung cancer metastasis. Methods: The expression of miRNAs in high metastatic lung cancer cell line (PGBE1) and low metastatic lung cancer cell line (PGLH7) was detected by non-coding microRNA microarray and identified by qPCR. Results: The results of non-coding microRNA microarray showed that four miRNAs differentially expressed in different metastatic lung cancer cells were hsa-mir-34b, hsa-mir-23a, hsa-mir-23b and hsa-mir-199a -5p. The differences of miRNAs were identified by real-time PCR. The abundance of 4 differential miRNAs in low metastatic cell line (PGLH7) was higher than that in high metastatic cell line (PGBE1) (P <0.05) The result is consistent. Conclusion: Four miRNAs may be involved in the metastasis of lung cancer. To investigate the miRNA function will be helpful to study the molecular mechanism of lung cancer metastasis.
