目的:观察加味四逆散对青少期应激大鼠行为学及海马、前额叶皮质中单胺类神经递质含量的影响。方法:将24只21日龄Wistar雄性大鼠随机分为空白组、模型组、氟西汀组、加味四逆散组;除空白组以外的3组予慢性温和不可预计性应激(CUMS)。造模结束后进行行为学观察,取海马及前额叶皮质,用高效液相-荧光法检测单胺类神经递质含量。结果:与空白对照组相比,模型组体质量、糖水偏爱度、旷场实验总路程、活动度、中央区路程,海马及前额叶皮质中单胺类神经递质含量均降低。加味四逆散组大鼠体质量、糖水偏爱度、旷场测试中活跃度、中央区活动路程、总路程均增加,海马及前额叶中单胺类神经递质含量均明显升高。结论:加味四逆散可能通过提高海马及前额叶皮质中单胺类神经递质的含量而改善CUMS的引起青少期大鼠抑郁样行为。
Objective: To observe the effects of Jiawei Sini Decoction on the behavior and the content of monoamine neurotransmitters in hippocampus and prefrontal cortex of adolescent stress rats. Methods: Twenty-four Wistar male rats of 21 days old were randomly divided into blank group, model group, fluoxetine group and Jiawei Sini Decoction group. Except for the blank group, three groups were given chronic mild unpredictable stress (CUMS) . After modeling, behavioral observation was performed. The hippocampus and prefrontal cortex were harvested and the content of monoamine neurotransmitters was measured by HPLC-Fluorometry. Results: Compared with the blank control group, body mass, glyco-water preference, open-field total distance, activity, central distance, monoamine neurotransmitters in the hippocampus and prefrontal cortex decreased in model group. The rats in Jiawei Sini San group had higher body weight, sugar preference, activity in open field test, distance traveled and distance traveled in the central area, and the contents of monoamine neurotransmitters in hippocampus and prefrontal lobe were significantly increased. CONCLUSION: Jiawei Sini San may improve CUMS induced depression-like behavior in adolescent rats by increasing monoamine neurotransmitters in hippocampus and prefrontal cortex.
