目的:研究葶苈子的DNA指纹图谱,初步探讨AFLP分子标记技术在葶苈子鉴别上的应用。方法:采用AFLP分子标记技术,对中药葶苈子及其常见混淆品荠菜和印度蔊菜进行基因组DNA多态性分析。结果与结论:选用16对EcoR I/MseI引物进行扩增,共检测到清晰条带1346条。其中多态性条带1303条,特异性条带759条,多态率达96.8%。每对引物组合中样品均扩增出自己的特征带,引物利用率为100%。丰富的特异性条带组合可作为样品鉴定的依据。此外,利用UP-GAM法构建样品间聚类图谱,聚类结果显示南、北葶苈子首先聚为葶苈子类,然后再和荠菜聚在一起,在遗传上两者有着较近的亲缘关系,印度蔊菜单独聚为一类。
OBJECTIVE: To study the DNA fingerprinting of 葶 苈 子 and to explore the application of AFLP molecular marker in the identification of 葶 苈 子. Methods: The AFLP molecular marker technique was used to analyze genomic DNA polymorphism of Chinese prickly heat and its common confused Shepherd’s purse and Indian pricklyaws. RESULTS AND CONCLUSION: Sixteen pairs of EcoR I / MseI primers were selected for amplification. A total of 1346 clear bands were detected. There were 1303 polymorphic bands and 759 specific bands with a polymorphism rate of 96.8%. Each pair of primer combinations amplified their own characteristic bands, primer utilization was 100%. Abundant specificity of the band combination can be used as a sample identification basis. In addition, the UP-GAM method was used to construct the inter-sample cluster map. The clustering results showed that S. przewalskis was first clustered in the razor clam, and then clustered together with shepherd’s purse, which had a relatively close genetic relationship Relationship, India 蔊 menu alone into a category.
