采用共价键合方法,制备了支化聚乙烯亚胺修饰的触须式聚合物开管柱,并对其电色谱性能展开了详细研究。开管柱的电渗流强度随着乙腈含量的升高而降低,随着流动相中缓冲液盐浓度的升高而减小。当流动相pH值低于7.5时,开管柱可获得反向电渗流;电渗流EOF日内、日间和周内的测定相对标准偏差RSD在0.8%—2.6%之间,柱间RSD为1.6%(n=4),批间RSD为4.8%(n=3),重现性较好。蛋白质在该柱上得到成功分离,没有严重拖尾。
The covalent bonding method was used to prepare a branched poly (ethyleneimine) modified whisker polymer column and its electrochromatographic properties were studied in detail. The electroosmotic flow intensity of open columns decreases with increasing acetonitrile content and decreases with increasing buffer salt concentration in the mobile phase. When the pH value of the mobile phase was less than 7.5, the reverse osmosis was obtained by open column. The relative standard deviations (RSDs) of EOF days, daytime and week were between 0.8% -2.6%, and the inter-column RSD was 1.6 % (n = 4). The inter-assay RSD was 4.8% (n = 3) with good reproducibility. The protein was successfully separated on the column without severe tailing.
