目的:研究Slug和E-cadherin在肾透明细胞癌(clear cell renal cell carcinoma,ccRCC)组织中的表达及其临床意义。方法:采用免疫组化SP法检测83例ccRCC组织和40例癌旁组织中Slug和E-cadherin蛋白的表达。采用Real-time PCR检测20例ccRCC及癌旁新鲜组织中Slug和E-cadherin mRNA的表达。分析二者蛋白和mRNA表达的相关性及Slug和E-cadherin蛋白表达与临床病理参数及预后的相关性。结果:Slug蛋白和mRNA在ccRCC组织中的表达明显高于癌旁组织。E-cadherin蛋白和mRNA在ccRCC组织中的表达明显低于癌旁组织(P<0.05)。ccRCC组织中Slug和E-cadherin蛋白和mRNA表达均具有显著负相关性(P<0.05)。Slug蛋白的表达与肿瘤大小、复发、TNM分期、淋巴结转移及血管侵袭密切相关(P<0.05);E-cadherin蛋白的表达与性别、肿瘤大小、TNM分期、淋巴结转移及血管侵袭密切相关(P<0.05)。Slug蛋白高表达的患者预后明显差于低表达的患者(P<0.05);而E-cadherin蛋白高表达的患者预后明显优于低表达的患者(P<0.05)。COX多因素回归分析显示Slug和E-cadherin蛋白表达状态是影响ccRCC患者预后的独立因素。结论:Slug和E-cadherin在ccRCC发生发展中发挥重要作用,可作为ccRCC预后判断的生物标志物。
Objective: To study the expression of Slug and E-cadherin in clear cell renal cell carcinoma (ccRCC) and its clinical significance. Methods: Immunohistochemical SP method was used to detect the expression of Slug and E-cadherin in 83 cases of ccRCC tissues and 40 cases of paracancerous tissues. Real-time PCR was used to detect the mRNA expression of Slug and E-cadherin in 20 cases of ccRCC and adjacent non-cancerous tissues. The correlation between protein and mRNA expression and the relationship between Slug and E-cadherin protein expression and clinical pathological parameters and prognosis were analyzed. Results: The expression of Slug protein and mRNA in ccRCC tissues was significantly higher than that in paracancerous tissues. The expression of E-cadherin protein and mRNA in ccRCC tissues was significantly lower than that in paracancerous tissues (P <0.05). There was a significant negative correlation between Slug and E-cadherin protein and mRNA expression in ccRCC (P <0.05). The expression of Slug protein was closely related to tumor size, recurrence, TNM stage, lymph node metastasis and vascular invasion (P <0.05). The expression of E-cadherin protein was closely related to gender, tumor size, TNM stage, lymph node metastasis and vascular invasion <0.05). The prognosis of patients with high expression of Slug protein was significantly lower than that of patients with low expression (P <0.05). The prognosis of patients with high E-cadherin protein expression was significantly higher than that of patients with low expression of Slug protein (P <0.05). COX multivariate regression analysis showed that Slug and E-cadherin protein expression status is an independent factor affecting the prognosis of patients with ccRCC. Conclusion: Slug and E-cadherin play an important role in the development of ccRCC and can be used as biomarkers for the prognosis of ccRCC.
