云南省流行病防治研究所病毒室用BALB/C小鼠骨髓瘤细胞(SP_2/O)与经基孔肯尼雅(CHIK)病毒免疫后的BALB/C小鼠的B淋巴细胞进行融合,共获得11株能分泌抗CHIK病毒MCAb杂交瘤细胞系。融合率为80.6%;阳性率为 29.7%。经微量中和试验证明11株MCAb均具有抗CHIK原型及从棕果蝠与人体分离到的地方株CHIK病毒的中和抗体。血凝抑制试验及免疫荧光均为阳性,MCAb培养上清效价前者为1:32—1:64,后者为1:40—1:80,MCAb腹水效价前者 1:2560—1:5120,后者1:2560—1:10240。和甲组虫媒病毒的马亚罗(MAY)及黄病毒中的乙脑(JE)、登革(DEN)、墨累山谷(MVE)、柯隆努尔森病毒(KFD)、兰加特(LGT)等均不发生交叉反应。11株MCAb经中国医学科学院医学生物研究所鉴定为IgG2b亚类。
Yunnan Provincial Institute of Epidemiology and Prevention BALB / C mouse myeloma cells (SP2 / O) were fused with B lymphocytes from BALB / c mice immunized with Chikungunya virus Eleven strains that can secrete anti-CHIK virus MCAb hybridoma cell line were obtained. The fusion rate was 80.6%; the positive rate was 29.7%. The results of micro-neutralization test showed that 11 strains of MCAb had the anti-CHIK prototype and the neutralizing antibody of CHIK virus isolated from the brown bats and the human body. The hemagglutination inhibition test and immunofluorescence were positive. The former titer of MCAb culture supernatant was 1: 32-1: 64, while the latter was 1:40-1: 80. The former titer of MCAb ascites 1: 2560-1: 5120 , The latter 1: 2560-1: 10240. (MAY) of group A arbovirus and JE, DEN, MVE, KFN, (LGT) do not cross-react. Eleven MCAbs were identified as IgG2b subclasses by the Institute of Medical Biology, Chinese Academy of Medical Sciences.
