目的研究腺苷A2a受体对肺泡上皮A549细胞钠离子通道α亚基(epithelial sodium channelα-subunit,α-ENaC)表达变化的影响,探讨其在急性肺损伤/急性呼吸窘迫综合征中的作用。方法不同剂量(0、0.1、1、10、100μmol/L)和不同时间(0、1、4、8、24、48h)的腺苷A2a受体激动剂CGS-21680干预A549细胞,分别用RT-PCR和Western blot检测α-ENaC mRNA和蛋白表达情况。结果①不同剂量CGS-21680作用A549细胞8h后,α-ENaC mRNA和蛋白表达水平从0.1μmol/L开始增加,并与CGS-21680剂量呈正相关(mRNA表达:r=0.959,P<0.01;蛋白表达:r=0.988,P<0.01),与对照组比较差异具有统计学意义(P<0.05)。②100μmol/LCGS-21680作用A549细胞1h后,α-ENaC mRNA和蛋白表达水平开始增加,4h后与对照组相比差异具有统计学意义(P<0.05)。结论激动腺苷A2a受体能上调α-ENaC的表达,有益于急性肺损伤/急性呼吸窘迫综合征的预后。
Objective To investigate the effect of adenosine A2a receptor on the expression of α-ENaC in alveolar epithelial A549 cells and its role in acute lung injury / acute respiratory distress syndrome. Methods A549 cells were treated with adenosine A2a receptor agonist CGS-21680 at different doses (0,0.1,1,10,100μmol / L) and different time (0,1,4,8,24,48h) -PCR and Western blot detection of α-ENaC mRNA and protein expression. Results ① The mRNA and protein expressions of α-ENaC in A549 cells after different doses of CGS-21680 were increased from 0.1μmol / L and positively correlated with the dose of CGS-21680 (r = 0.959, P <0.01) Expression: r = 0.988, P <0.01), the difference was statistically significant compared with the control group (P <0.05). After treated with 100μmol / L of LCGS-21680 for 1h, the expression of α-ENaC mRNA and protein began to increase, and the difference was statistically significant after 4h compared with the control group (P <0.05). Conclusions The activation of adenosine A2a receptor can upregulate the expression of α-ENaC, which is beneficial to the prognosis of acute lung injury / acute respiratory distress syndrome.
