目的高效液相色谱法(HPLC法)测定阿图什无花果叶汁中伞形花内酯、佛手柑内酯和补骨脂素3种香豆素的含量。方法色谱柱Sunfire C_(18)ODS(250 mm×4.6 mm,5μm),流速1 mL、min,检测波长为220 nm,柱温30℃,流动相为乙腈-水,流动相配比为0~10 min,乙腈25%;10~12min,乙腈由25%梯度增至50%;12~20 min,乙腈50%;20~22 min,乙腈由50%梯度增至100%。结果伞形花内酯、佛手柑内酯和补骨脂素分别在0.001~0.019 mg、mL(R~2=0.999 9)、0.01~0.19 mg、mL(R~2=0.9997)和0.01~0.19 mg、mL(R~2=0.9999)范围内与峰面积呈良好线性关系,平均回收率分别为98.09%(RSD=0.85%)、102.07%(RSD=1.07%)和101.00%(RSD=0.71%)。阿图什无花果叶汁中伞形花内酯、佛手柑内酯和补骨脂素3种香豆素的含量分别为1.09、9.89、24.58 mg/g。结论HPLC法简便、准确、稳定,可用于测定无花果叶汁中伞形花内酯、佛手柑内酯和补骨脂素的含量。
OBJECTIVE To determine the contents of three kinds of coumarin in Undaria pintail fig leaves by HPLC (HPLC method). Methods The column was Sunfire C 18 ODS (250 mm × 4.6 mm, 5 μm) with a flow rate of 1 mL and min. The detection wavelength was 220 nm and the column temperature was 30 ℃. The mobile phase consisted of acetonitrile-water with a mobile phase ratio of 0-10 min, acetonitrile 25%; 10-12 min, acetonitrile 25% gradient to 50%; 12 ~ 20 min, acetonitrile 50%; 20 ~ 22 min, acetonitrile gradient from 50% to 100%. Results The results showed that ursovalerate, bergapten and psoralen were in the range of 0.001-0.019 mg, mL (R 2 = 0.999 9), 0.01-0.19 mg, mL (R 2 = 0.9997) and 0.01-0.19 The average recoveries were 98.09% (RSD = 0.85%), 102.07% (RSD = 1.07%) and 101.00% (RSD = 0.71% ). The contents of umbelliferone, bergapten and psoralen in the leaves of Atushi fig were 1.09, 9.89 and 24.58 mg / g, respectively. Conclusion HPLC method is simple, accurate and stable. It can be used to determine ursolumin, bergamot and psoralen in fig leaf juice.
