目的探讨血卟啉单甲醚(HMME)和声诺维(SonoVue)在超声辐照下对MDA-MB-231细胞凋亡的影响。方法将处于对数生长期的MDA-MB-231细胞分为5组(对照组、U、UM、UH组和UHM组)。采用优化后的实验参数,50Hz脉冲波照射60s。采用流式细胞仪检测照射后MDA-MB-231细胞的线粒体膜电位和凋亡率。结果对照组、U、UH、UM、UHM组的几何平均荧光强度值分别为(85.1±17.3)、(89.0±13.8)、(146.4±12.8)、(97.3±23.2)、(154.8±19.6)。对照组、U、UH、UM、UHM组的细胞凋亡率分别为(1.62±0.49)%、(14.16±3.34)%、(26.03±5.79)%、(53.08±12.71)%、(69.53±9.79)%,经过N-乙酰半胱氨酸(NAC)预处理后,UH+NAC组、UM+NAC组、UHM+NAC组的细胞凋亡率分别为(14.15±4.17)%、(24.36±6.66)%、(47.21±5.00)%。结论 SonoVue和HMME在超声辐照下能够相互协同诱导MDA-MB-231细胞凋亡,活性氧参与该过程。
Objective To investigate the effects of hematoporphyrin monomethyl ether (HMME) and SonoVue on the apoptosis of MDA-MB-231 cells under ultrasound irradiation. Methods MDA-MB-231 cells in logarithmic growth phase were divided into 5 groups (control group, U, UM, UH group and UHM group). Optimized experimental parameters, 50Hz pulse wave irradiation 60s. The mitochondrial membrane potential and apoptosis rate of MDA-MB-231 cells were detected by flow cytometry. Results The geometric mean fluorescence intensities of the control group, U, UH, UM and UHM groups were (85.1 ± 17.3), (89.0 ± 13.8), (146.4 ± 12.8), (97.3 ± 23.2) and (154.8 ± 19.6), respectively. The apoptotic rates in the control group, U, UH, UM and UHM groups were (1.62 ± 0.49)%, (14.16 ± 3.34)%, (26.03 ± 5.79)%, (53.08 ± 12.71)% and (69.53 ± 9.79 ), The apoptosis rates of UH + NAC group, UM + NAC group and UHM + NAC group were (14.15 ± 4.17)% and (24.36 ± 6.66) after NAC pretreatment respectively )%, (47.21 ± 5.00)%. Conclusions SonoVue and HMME can synergistically induce the apoptosis of MDA-MB-231 cells under the action of ultrasound irradiation, and reactive oxygen species participate in this process.
