建立以离子液体为萃取剂,结合超声辅助萃取,利用高效液相色谱法测定垂丝海棠叶中根皮苷、3-羟基根皮苷和槲皮苷含量的方法。采用Agilent TC-C18柱(4.6 mm×250 mm,5μm)进行分离,以乙腈~(-1)%磷酸水溶液(20∶80)为流动相,紫外检测波长为270 nm,流速0.8 m L·min~(-1),柱温为室温。在优化条件下,根皮苷、3-羟基根皮苷、槲皮苷在0.9~112.5μg(r=0.999 6),0.093 2~11.65μg(r=0.999 1),0.097 2~12.15μg(r=0.999 8)线性关系良好,平均回收率分别为99.35%,98.80%,98.19%。该方法绿色环保、快速简便、具有良好的重复性,适用于同时测定垂丝海棠叶中根皮苷、3-羟基根皮苷和槲皮苷的含量。
A method for determining the content of phlorizin, 3-hydroxyprophyloside and quercitrin in the leaves of Begonia spinosta by ionic liquid as extractant combined with ultrasonic-assisted extraction and high performance liquid chromatography was established. The separation was performed on an Agilent TC-C18 column (4.6 mm × 250 mm, 5 μm) using acetonitrile-1% phosphoric acid (20:80) as the mobile phase at UV detection wavelength of 270 nm and flow rate of 0.8 mL · min ~ (-1), the column temperature is room temperature. Under the optimized conditions, the contents of phlorizin, 3-hydroxypendothelin and quercitrin in the range of 0.9-112.5 μg (r = 0.999 6), 0.093 2-11.65 μg (r = 0.999 1), 0.097 2-12.15 μg = 0.999 8). The average recoveries were 99.35%, 98.80% and 98.19%, respectively. The method is environment-friendly, fast and simple, and has good repeatability. The method is suitable for simultaneous determination of phlorizin, 3-hydroxy phlorizin and quercitrin in leaves of Begonia spinosa.
