目的研究雷公藤内酯醇(triptolide,T10)对β-淀粉样蛋白(β-amyloid,Aβ)1-42激活的小胶质细胞一氧化氮(nitrogenmonoxidum,NO)的产生及诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)表达的影响。方法用不同浓度的T10(10-11、10-10、10-9、10-8mol/L)预孵育小胶质细胞12 h,然后加入10μmol/L Aβ1-42共孵育12 h;Griess反应检测上清NO的含量,Western blotting检测iNOS蛋白的表达。结果 Aβ组iNOS的表达和NO释放明显高于对照组(P<0.01)。T10可明显减少Aβ诱导的小胶质细胞iNOS的表达及NO的释放(P<0.05)。结论 T10可抑制Aβ1-42激活的小胶质细胞iNOS蛋白的表达及NO的释放。
Objective To investigate the effects of triptolide (T10) on the production of nitric oxide (NO) and the inducible nitric oxide synthase (NOS) in microglia activated by β-amyloid (Aβ) 1-42 (Inducible nitric oxide synthase, iNOS) expression. Methods Microglia were preincubated with different concentrations of T10 (10-11,10-10,10-9,10-8 mol / L) for 12 h, then incubated with 10 μmol / L Aβ1-42 for 12 h. The Griess reaction The content of NO in supernatant and the expression of iNOS protein were detected by Western blotting. Results The expression of iNOS and NO release in Aβ group were significantly higher than those in control group (P <0.01). T10 can significantly reduce Aβ-induced microglial iNOS expression and NO release (P <0.05). Conclusion T10 can inhibit the expression of iNOS protein and the release of NO in Aβ1-42-activated microglia.
