目的 :探讨CⅡ诱导的RA患者关节滑液中自身反应性T细胞免疫学特性。方法 :分离RA患者关节滑液中SFMC ,用CⅡ刺激后分析反应性的T细胞频率 ;用ELISA方法检测CⅡ刺激SFMC后上清中的IFN γ和IL 4的含量及通过流式细胞检测SFMC表面标志的表达 ;应用半定量PCR分析外周血及滑膜液中TCRVβ基因表达格局。 结果 :SFMC中CⅡ反应性的T细胞频率高于PBL ;IFN γ的含量在CⅡ刺激SFMC后上清中较高 ,而在未刺激中较低 (P <0 0 1) ,在PBL中更低 (P <0 0 5 ) ;SFMC上CD8及CXCR3表达多于PBMC(P <0 0 5 ) ;外周血T细胞表达绝大多数Vβ基因 ,而滑膜液中TCRVβ基因取用呈现明显的不均衡性。结论 :CⅡ反应性T细胞在SFMC中占优势 ,可能是介导RA发病的主要原因
Objective: To investigate the immunological characteristics of autoreactive T cells in synovial fluid of C Ⅱ -induced RA patients. Methods: SFMC in synovial fluid from patients with RA were isolated and the frequency of reactive T cells was analyzed after stimulated with CⅡ. The contents of IFNγ and IL-4 in supernatant of CⅡ-stimulated SFMC were detected by ELISA. The surface of SFMC was detected by flow cytometry The expression of TCRVβgene in peripheral blood and synovial fluid was analyzed by semi-quantitative PCR. Results: The frequency of CⅡreactive T cells in SFMC was higher than that in PBL. The content of IFNγ was higher in supernatant of CⅡ-stimulated SFMC but lower in unstimulated (P <0.01) and lower in PBL (P <0.05). The expression of CD8 and CXCR3 in SFMC was more than that in PBMC (P <0.05). The majority of peripheral blood T cells expressed Vβ gene, but the TCRVβ gene in synovial fluid showed obvious imbalance Sex. Conclusion: C Ⅱ reactive T cells predominate in SFMC and may be the main reason for the pathogenesis of RA
