目的探讨嵌合有免疫刺激分子CD40配体(CD40L)或粒细胞巨噬细胞集落刺激因子(GM-CSF)的汉滩病毒(HTNV)样颗粒(VLP)对小鼠骨髓细胞活化的影响,以确定嵌合汉滩病毒样颗粒的生物学活性。方法利用真核细胞瞬时表达VLP、VLP-CD40L、VLP-GM-CSF,纯化后与小鼠骨髓细胞共培养,通过流式细胞术检测小鼠骨髓细胞活化情况。结果三种汉滩病毒VLP均能促进小鼠骨髓细胞活化(P<0.05),其中嵌合有GM-CSF因子的VLP促进小鼠骨髓细胞活化更为明显(P<0.01)。结论真核细胞表达的三种汉滩病毒VLP均有一定的生物学活性,为进一步研究汉滩病毒VLP疫苗奠定基础。
Objective To investigate the effect of HTNV-like particles (VLP) chimeric with CD40L or GM-CSF on the activation of bone marrow cells in mice Determine the biological activity of chimeric Hantaan virus-like particles. Methods VLP, VLP-CD40L and VLP-GM-CSF were transiently expressed in eukaryotic cells. After purification, they were co-cultured with mouse bone marrow cells. The activation of mouse bone marrow cells was detected by flow cytometry. Results All three Hantaan virus VLPs could promote the activation of bone marrow cells in mice (P <0.05). The VLPs chimeric with GM-CSF promoted the activation of bone marrow cells in mice (P <0.01). Conclusion All of the three Hantaan virus VLPs expressed in eukaryotic cells have certain biological activity, which lays the foundation for further study of the Hantaan virus VLP vaccine.
