目的 :研究正常人骨髓基质祖细胞 ( CFU- F)在体外传代培养过程中的功能变化。方法 :采用体外液体单层和半固体培养法。结果 :F1传代细胞对骨髓红系祖细胞集落形成单位 ( CFU- E)和粒系祖细胞集落形成单位 ( CFU- GM)支持能力最显著 ,F2 代对有核细胞粘附能力最显著。结论 :F1代是进行第二次接种骨髓细胞的最佳时机 ,而对有核细胞的粘附能力主要与成纤维细胞有关。
Objective: To study the functional changes of normal human bone marrow stromal cells (CFU-F) during subculture. Methods: In vitro liquid monolayer and semi-solid culture method. Results: F1 passaged cells had the most significant ability to support CFU-E and CFU-GM of myeloid erythroid progenitor cells, and F2 cells had the most significant adhesion ability to nucleated cells. Conclusion: F1 generation is the best time for the second inoculation of myeloid cells, while the adhesion to nucleated cells is mainly related to fibroblasts.
