目的探讨补肾养肝熄风汤对帕金森病(PD)小鼠纹状体神经递质和多巴胺(DA)受体的调节机制。方法将C57/BL雄性小鼠56只随机分为正常组、模型组、中药组和西药组,每组14只。采用MPTP腹腔注射复制PD小鼠模型,正常组、模型组生理盐水灌胃(0.1ml/10g),中药组以补肾养肝熄风汤灌胃(0.1ml/10g),西药组多巴丝肼灌胃(0.1ml/10g),每日1次,治疗14d。治疗结束后分离取出小鼠纹状体组织,测定纹状体DA、去甲肾上腺素(NE)、5-羟色胺(5-HT)、亮胺酸脑啡肽(LEK)含量以及DAD1、D2受体的含量。结果模型组、中药组和西药组小鼠纹状体DA、NE、5-HT、LEK含量显著低于正常组(P<0.05或P<0.01),中药组和西药组DA、NE、5-HT含量显著高于模型组(P<0.05或P<0.01);模型组、中药组和西药组纹状体DAD1受体免疫组化指数(IHC)显著低于正常组(P<0.01),西药组DAD1受体IHC高于模型组和中药组(P<0.05);中药组、西药组DAD2受体IHC显著高于模型组(P<0.01)。结论补肾养肝熄风汤可提高纹状体下降的DA、NE、5-HT水平,可提高纹状体DAD2受体的含量。
Objective To investigate the regulatory mechanism of Bushen Yanggan Xifeng Decoction on neurotransmitter and dopamine (DA) receptors in striatum of Parkinson’s disease (PD) mice. Methods 56 C57 / BL male mice were randomly divided into normal group, model group, traditional Chinese medicine group and western medicine group, with 14 in each group. The model of PD mice was injected intraperitoneally with MPTP. Normal group and model group were given normal saline (0.1ml / 10g) Gavage (0.1ml / 10g), 1 day, treatment 14d. After treatment, the striatum DA, norepinephrine (NE), serotonin (5-HT), leucine enkephalin (LEK) content and DAD1, D2 Body content. Results The contents of DA, NE, 5-HT and LEK in the model group, the traditional Chinese medicine group and the western medicine group were significantly lower than those in the normal group (P <0.05 or P <0.01) (P <0.05 or P <0.01). The immunohistochemical index of DAD1 receptor in striatum of model group, traditional Chinese medicine group and western medicine group was significantly lower than that of normal group (P <0.01), Western medicine The IHC of DAD1 receptor in group D was higher than that in model group and Chinese medicine group (P <0.05). IHC of DAD2 receptor in Chinese medicine group and western medicine group was significantly higher than that in model group (P <0.01). Conclusion Bushen Yanggan Xifeng Decoction can increase the levels of DA, NE and 5-HT in striatum and increase the content of DAD2 receptors in the striatum.
