目的:研究白首乌C21甾总苷醋酸水解的最佳条件。方法:以开德苷元-3-O-β-D-洋地黄毒糖苷、告达庭、开德苷元-3-O-α-L-迪吉糖-(1→4)-β-D-磁麻糖苷和告达庭-3-O-β-D-磁麻糖苷的含量为指标,采用正交试验设计L9(33)进行优选,考察水解温度(A)、水解时间(B)、醋酸浓度(C)3个因素对白首乌C21甾总苷醋酸水解条件的影响,同时建立HPLC含量测定的方法。结果:影响醋酸水解的主次因素为A>B>C,最佳水解条件为5.0%醋酸溶液,100℃回流6 h。结论:此水解条件明显提高了白首乌中抗癌有效成分的含量,对研究抗肿瘤药物具有重要作用。
Objective: To study the optimum conditions for the hydrolysis of C21-steroidal glycosides in Radix Methods: The cells were cultured with orotaxel-3-O-β-D-digoxigenin, D-Magnatioside and Dota Ting-3-O-β-D-Magnatioside were used as indexes to optimize L9 (33). The hydrolysis temperature (A), hydrolysis time (B) , Acetic acid concentration (C) on the hydrolysis of C21-STAD in acetic acid and the determination of HPLC. Results: The primary and secondary factors affecting acetic acid hydrolysis were A> B> C, the optimal hydrolysis conditions were 5.0% acetic acid solution and refluxed at 100 ℃ for 6 h. Conclusion: The hydrolysis conditions significantly increased the anti-cancer active ingredients in Radix Polygoni Multiflori, which plays an important role in the study of anti-tumor drugs.
