目的:研究不同浓度苦参碱对生物膜状态下的白色念珠菌ALS基因mRNA表达量的影响。方法:采用白色念珠菌标准菌株ATCC76615为研究对象,经载玻片法形成白色念珠菌生物膜。在经不同浓度苦参碱干预下,通过RT-PCR法检测生物膜状态白色念珠菌ALS1/ALS4基因mRNA表达量的变化。结果:不同浓度苦参碱干预下生物膜状态白色念珠菌黏附相关基因ALS1/ALS4 mRNA表达减弱。1~8 mg/m L苦参碱干预下ALS1/ALS4的mRNA表达相对量分别为0.96~0.34/0.93~0.28。结论:苦参碱可抑制白色念珠菌粘附相关基因ALS1/ALS4 mRNA水平的表达。
Objective: To study the effects of different concentrations of matrine on the expression of albicans albicans ALS mRNA in biofilm. Methods: Candida albicans standard strain ATCC76615 was used as the research object, and Candida albicans biofilm was formed by slide method. The changes of ALS1 / ALS4 gene mRNA expression in the biofilm state were detected by RT-PCR under the different concentrations of matrine. Results: The expression of ALS1 / ALS4 mRNA of Candida albicans adhesion-related genes in biofilm-conditioned medium with different concentrations of matrine was weakened. The relative mRNA levels of ALS1 / ALS4 in the matrine range of 1 ~ 8 mg / m L were 0.96 ~ 0.34 / 0.93 ~ 0.28, respectively. Conclusion: Matrine can inhibit the expression of ALS1 / ALS4 mRNA in Candida albicans adhesion-related genes.
