目的了解吉林省麻疹病毒流行株的基因型别,探讨控制麻疹发生的措施。方法在2005年1—12月采集吉林省麻疹暴发和散发病人的咽喉拭子和尿液标本共72份,用 CDW_(150)细胞分离到麻疹病毒9株。用逆转录-聚合酶链反应(RT-PCR)从麻疹病毒中扩增出核蛋白(N)基因羧基端450个核苷酸片段,进行核苷酸序列分析和基因分型。结果 9株麻疹病毒均属 H_1基因型,其中H1a7株,H1b2株,H1a 和 H1b 在450个核苷酸片段中的差异为2.0%~3.5%。结论 H_1基因型是吉林省麻疹病毒的优势流行株,是导致2005年吉林省麻疹局部暴发和散发的主要病原体。
Objective To understand the genotypes of measles virus strains in Jilin Province and to explore the measures to control the occurrence of measles. Methods A total of 72 swabs and urine samples of measles outbreaks and sporadic cases were collected from January to December 2005 in Jilin Province. Nine measles viruses were isolated from CDW 150 cells. Nucleotide 450 nucleotides fragment of nucleoprotein (N) gene was amplified from the measles virus by reverse transcription-polymerase chain reaction (RT-PCR), and the nucleotide sequence and genotyping were performed. Results All the 9 measles viruses belonged to genotype H_1, of which H1a7, H1b2, H1a and H1b varied from 2.0% to 3.5% in 450 nucleotide fragments. Conclusion H_1 genotype is the predominant epidemic strain of measles virus in Jilin Province and is the major causative agent of local outbreak and spread of measles in Jilin Province in 2005.
