本文建立了一种简便的非放射性底物测定家兔组织H-TGL(肝脂酶)活性的方法。其最适条件为0.05 mol Tris-HCl,pH8.5缓冲体系,37℃保温30min。该酶活性可被50 mmol SDS完全抑制,而不受1 mol NaGl和鱼精蛋白抑制。喂胆固醇食料家兔的组织H-TGL活性明显低于正常家兔,其活性与血浆总胆固醇水平及ApoB水平呈明显直线负相关。体外实验证明,高胆固醇血症家兔的血浆在体外对H-TGL活性有明显抑制作用。本文并对H-TGL在脂蛋白代谢中的作用进行了讨论。
In this paper, a simple non-radioactive substrate was established to determine the activity of H-TGL (hepatic lipase) in rabbits. The optimal condition was 0.05 mol Tris-HCl, pH8.5 buffer system, incubated at 37°C for 30 min. The enzyme activity was completely inhibited by 50 mmol SDS, but not by 1 mol NaGl and protamine. The activity of H-TGL in rabbits fed with cholesterol diets was significantly lower than that in normal rabbits. The activity was significantly and negatively correlated with plasma total cholesterol and ApoB levels. In vitro experiments demonstrated that the plasma of hypercholesterolemic rabbits significantly inhibited H-TGL activity in vitro. This article discusses the role of H-TGL in lipoprotein metabolism.
