目的:观察吉西他滨在体外对人胚肾细胞HEK293和人乳腺癌细胞MCF-7增殖的影响。方法:采用MTT法测定吉西他滨对人胚肾细胞HEK293和人乳腺癌细胞MCF-7增殖的影响,比较两者IC50值,绘制细胞生长曲线;并测定吉西他滨作用后细胞培养上清液中SOD,MDA,LDH值的变化。结果:吉西他滨在体外对人乳腺癌细胞MCF-7的增殖抑制作用较强,同时对人胚肾细胞HEK293的增殖有一定的抑制作用,两者的化疗指数为分别为26.907(24h),15.307(48h),10.264(72h)。结论:吉西他滨对人乳腺癌细胞MCF-7有明显增殖抑制作用,同时可抑制人胚肾细胞HEK293的增殖,对肾脏有一定的毒性。
Objective: To observe the effect of gemcitabine on the proliferation of human embryonic kidney cell line HEK293 and human breast cancer cell line MCF-7 in vitro. Methods: The effects of gemcitabine on the proliferation of human embryonic kidney cell line HEK293 and human breast cancer cell line MCF-7 were determined by MTT assay. The cell growth curve was drawn by comparing the IC50 values of both, and the activities of superoxide dismutase , LDH value changes. Results: Gemcitabine inhibited the proliferation of human breast cancer cell line MCF-7 in vitro and inhibited the proliferation of human embryonic kidney cell line HEK293 in vitro. The chemotherapy index of gemcitabine was 26.907 (24h) and 15.307 48h), 10.264 (72h). Conclusion: Gemcitabine can significantly inhibit the proliferation of human breast cancer cell line MCF-7 and inhibit the proliferation of human embryonic kidney cell line HEK293, which is toxic to the kidney.
