采用5种诱导培养基和3种分化培养基对4个野生稻种与9个栽培稻品种组配的杂种后代进行花药培养,结果筛选出2种较好的诱导培养基:Ⅰ.籼稻培养基,并以水解乳蛋白300mg/l代替水解酪蛋白,甘氨酸2mg/l代替脯氨酸100mg/l;Ⅴ.培养基Ⅰ铁盐加倍,同时表明铁盐加倍对诱导愈伤无显著作用。筛选出1种较好的分化培养基:D3.MS培养基+KT2+NAA0.5+水解乳蛋白800+肌醇500(mg/l)。结果还表明不同基因型的野栽杂种花培愈伤诱导率和绿苗生产力差异很大
Five kinds of induction medium and three kinds of differentiation medium were used for anther culture of hybrid progeny of 4 wild rice varieties and 9 cultivated rice varieties. Two kinds of better induction mediums were screened out: Ⅰ. Indica medium, hydrolyzed casein with hydrolyzed milk protein 300mg / l, glycine 2mg / l instead of proline 100mg / l; Iron I doubled in culture medium Ⅰ, and at the same time it showed that doubling iron salts had no significant effect on inducing callus. Screen a better differentiation medium: D3. MS medium + KT2 + NAA0.5 + hydrolyzed milk protein 800 + myo-inositol 500 (mg / l). The results also show that different genotypes of wild-type hybrid flower callus induction rate and green seedling productivity vary greatly
