目的利用抗鼠疫噬菌体血清分离携带野生型鼠疫噬菌体的鼠疫菌。方法在鼠疫菌与鼠疫噬菌体并存的情况下,在赫氏琼脂培养基上先滴加抗鼠疫噬菌体血清,然后将被检材料三段划线接种于该平皿上,经培养后即可获得正常鼠疫菌。结果使用抗鼠疫噬菌体血清从携带有野生型鼠疫噬菌体的鼠疫菌中分离出正常的鼠疫菌落,该菌落无噬菌迹象,并被鼠疫噬菌体所裂解。结论利用抗鼠疫噬菌体血清分离携带野生型鼠疫噬菌体的鼠疫菌方便、特异、实用,同时对鼠疫检测、疫情判定、实验室保存菌株都将具有重要意义。
Objective To isolate Yersinia pestis carrying wild-type plague phage using anti-plague phage serum. Methods In the co-existence of plague bacteriophages and plague phage, Hep agar culture medium first dropping anti-plague phage serum, and then seized materials were streaked three-stranded on the plate, the culture can be obtained after normal plague bacteria. As a result, normal plague colonies were isolated from the plague blight carrying the phage of the wild-type plague using the anti-plague phage sera and the colonies showed no signs of phage and were lysed by the plague phage. Conclusion It is convenient, specific and practical to use the anti-plague phage serum to isolate the plague bacteriophage carrying the wild-type plague bacterioplankton. At the same time, it will be of great significance for the plague detection, epidemic situation determination and laboratory preservation of the strains.
