目的研究转铁蛋白与Bel-7402肝癌细胞耐药性的关系。方法根据已知转铁蛋白序列设计引物扩增基因片段,荧光定量PCR检测其在敏感Bel-7402细胞株、阿霉素抗性Bel-7402细胞株之间的表达水平差异。结果成功扩增Bel-7402肝癌细胞转铁蛋白基因片段,荧光定量PCR表明转铁蛋白基因在阿霉素抗性Bel-7402细胞株的表达量是敏感Bel-7402细胞株4.83倍。结论转铁蛋白基因对肝癌细胞的耐药发生发展存在一定的作用,可为研究肝癌耐药性的备选基因,以及为肝癌耐药性检测提供新的靶标。
Objective To study the relationship between transferrin and drug resistance of Bel-7402 hepatoma cells. Methods The primers were designed according to the known transferrin sequence to amplify the gene fragment. The difference of expression level between Bel-7402 cell line and Bel-7402 cell line was detected by fluorescence quantitative PCR. Results The fragment of transferrin gene of Bel-7402 hepatoma cells was successfully amplified. Fluorescence quantitative PCR showed that the expression level of transferrin gene in Bel-7402 cells transfected with doxorubicin was 4.83-fold more than that of Bel-7402 cells. Conclusion The transferrin gene may play a role in the development of drug resistance of hepatocellular carcinoma cells, which may provide a new target for studying the drug resistance of hepatocellular carcinoma and detecting the drug resistance of hepatocellular carcinoma.
