目的探讨E-cad、β-cat蛋白在神经母细胞瘤中的表达及其与该肿瘤特征和预后的关系。方法用免疫组化Envision法检测90例神经母细胞瘤瘤组织E-cad、β-cat蛋白的表达,结合随访资料进行统计学分析。结果①FH型β-cat的阳性率(67.1%)显著高于UFH型(40.0%)(P=0.028);②神经母细胞瘤β-cat异常表达率(100%)显著高于节细胞神经母细胞瘤(89.3%)(P=0.009);③E-cad的阳性表达率和异常表达率与肿瘤的组织学分型具有显著相关性;④肿瘤的分期和病理分型对生存时间有显著意义;⑤E-cad、β-cat的阳性表达率和异常表达率均与有无淋巴结转移显著相关。结论E-cad/β-cat复合体对维持细胞间粘附有重要作用,其结构异常和功能紊乱可促进肿瘤的浸润和转移,联合检测粘附分子E-cad、β-cat可作为NB侵袭、转移能力和分化程度等预后因素相关的指标,可对神经母细胞瘤的临床预后进行综合评估。
Objective To investigate the expression of E-cad, β-cat protein in neuroblastoma and its relationship with the characteristics and prognosis of the tumor. Methods The expression of E-cad and β-cat protein in 90 cases of neuroblastoma were detected by immunohistochemical Envision method. The follow-up data were used for statistical analysis. Results ① The positive rate of β-cat in FH was 67.1%, which was significantly higher than that in UFH (40.0%) (P = 0.028). ② The abnormal expression rate of β-cat in neuroblastoma was significantly higher than that in neurons (89.3%) (P = 0.009). The positive expression rate of E-cad and the abnormal expression rate were significantly correlated with the histological type of the tumor. The staging and pathological classification of the tumor had significant significance on the survival time. ⑤E -cad, β-cat positive expression rate and abnormal expression rate with or without lymph node metastasis were significantly correlated. Conclusions The E-cad / β-cat complex plays an important role in maintaining cell adhesion. Its structural abnormalities and dysfunction can promote tumor invasion and metastasis. Combined detection of E-cad and β-cat adhesion can be used as NB invasion , Metastasis ability and degree of differentiation and other prognostic factors related indicators, the clinical prognosis of neuroblastoma comprehensive assessment.
