目的建立一种前处理简单、灵敏度高、重现性好的馒头中合成色素的检测方法。方法采用高效液相色谱-二极管阵列检测(HPLC-DAD)法,样品用无水乙醇+氨水+水(7+2+1)溶液提取,Agilent HC-C18柱(250mm×4.6mm,5μm)液相色谱柱分离,乙酸铵缓冲溶液(0.02mol/L)-甲醇为流动相进行梯度洗脱,检测波长448nm。结果测定结果具有良好的线性和重复性(RSD:1.25%~2.85%),样品加标回收率为87.4%~90.3%,方法检出限为0.002g/kg。结论该方法具有简便、快速、准确等优点,可用于准确测定馒头中的合成色素。
OBJECTIVE To establish a method for the detection of synthetic pigment in steamed bread with simple pretreatment, high sensitivity and reproducibility. Methods The HPLC-DAD method was used. The samples were extracted with anhydrous ethanol + ammonia + water (7 + 2 + 1). The samples were separated on an Agilent HC-C18 column (250 mm × 4.6 mm, 5 μm) Separation of the column, ammonium acetate buffer solution (0.02mol / L) - methanol as the mobile phase gradient elution, detection wavelength 448nm. The results showed good linearity and reproducibility (RSD: 1.25% ~ 2.85%). The spiked recoveries were 87.4% ~ 90.3%. The detection limit was 0.002g / kg. Conclusion The method is simple, rapid and accurate, which can be used to accurately determine the synthetic pigment in steamed bread.
