目的:研究九龙藤总黄酮(Bauhinia championii Benth flavones,BCF)对大鼠心肌缺血再灌注损伤后自由基代谢及细胞凋亡的影响。方法:将实验动物随机分为6组:假手术组、心肌缺血再灌注模型组、九龙藤总黄酮(30、60、120和240mg/kg)预处理组、复方丹参滴丸(135mg/kg)阳性对照组,术前1周开始灌胃给药。测定各组大鼠血清中谷草转氨酶(AST)、肌酸激酶(CK)、乳酸脱氢酶(LDH)活性、丙二醛(MDA)含量以及总抗氧化能力(T-AOC);检测心肌组织中超氧化物歧化酶(SOD)、谷胱甘肽过氧化物酶(GSH-Px)、过氧化氢酶(CAT)活性以及丙二醛(MDA)含量;通过HE染色方法观察各组大鼠心肌组织病理学改变,并通过TUNEL染色观察细胞凋亡状况。结果:与模型对照组相比,九龙藤总黄酮(60、120和240mg/kg)预处理组大鼠血清中AST、CK、LDH活性显著降低,MDA含量显著升高,T-AOC显著升高;九龙藤总黄酮(60、120和240mg/kg)预处理组心肌组织中SOD、GSH-Px、CAT活性显著升高、MDA含量显著降低;心肌组织病理性改变和细胞凋亡程度均显著较轻。其中以240mg/kg组各指标变化最显著。结论:九龙藤总黄酮能够有效增强抗氧化酶活性,提高自由基清除能力,改善组织病理学改变,抑制细胞凋亡,提示九龙藤总黄酮对大鼠心肌缺血再灌注损伤具有剂量依赖性的保护作用。
Objective: To investigate the effects of total flavonoids (BCh) from Kowloon vine on the metabolism of free radicals and apoptosis after myocardial ischemia / reperfusion injury in rats. Methods: The experimental animals were randomly divided into 6 groups: sham operation group, model group of myocardial ischemia-reperfusion, pretreatment group of Jiulongtong total flavonoids (30,60,120 and 240mg / kg), compound Danshen dropping pills (135mg / kg ) Positive control group, one week before surgery started gavage. Results: Compared with the model control group, the activities of AST, CK and LDH in the serum of total flavonoids of Jiulongtong (60, 120 and 240 mg / kg) were significantly decreased, the content of MDA was significantly increased and T-AOC was significantly increased ; The activities of SOD, GSH-Px and CAT in myocardium of pretreated group (60, 120 and 240 mg / kg) were significantly increased, the content of MDA was significantly decreased; the pathological changes of myocardial tissue and the degree of apoptosis were significantly light. Among them, 240mg / kg group showed the most significant change. CONCLUSION: The total flavonoids of Jiulongtang can effectively enhance the activity of antioxidant enzymes, improve the scavenging ability of free radicals, improve the histopathological changes and inhibit the apoptosis of the cells. It is suggested that the total flavonoids of Jiulongtong can inhibit the myocardial ischemia-reperfusion injury in a dose-dependent manner Protective effects.
