目的研究克班宁脂质体的处方筛选和制备工艺。方法采用p H值梯度法制备克班宁脂质体,以包封率和载药量为评价指标,采用葡聚糖凝胶过滤法分离脂质体,紫外分光光度法测定克班宁含量,采用正交设计优化制备工艺。结果最佳工艺为:药脂比为1∶10、胆固醇与磷脂比为1∶5,50℃恒温水浴孵育20 min,制备克班宁脂质体。结论 p H值梯度法制备的克班宁脂质体处方合理,工艺可行,包封率较高。
Objective To study the formulation and preparation of Kebaning liposomes. Methods The Klebsiella liposomes were prepared by p H value gradient method. The entrapment efficiency and drug loading were used as the evaluation indexes. The liposomes were separated by Sephadex. The contents of Kebaning were determined by ultraviolet spectrophotometry. Orthogonal design to optimize the preparation process. Results The optimal process was as follows: Lipid ratio 1:10, Cholesterol and phospholipid ratio 1: 5, 50 ℃ constant temperature water bath for 20 min to prepare Kebaning liposomes. Conclusions Kebaning liposome prepared by p H value gradient method has reasonable prescription, feasible technology and high entrapment efficiency.
