本试验旨在单因素试验的基础上,考察乙醇体积分数、料液比、微波处理时间对车前子黄酮提取率的影响,利用响应面法确定微波提取车前子黄酮的最佳工艺,并研究微波提取的车前子黄酮对环磷酰胺(cyclophosphamide,CY)诱导鸡氧化应激的影响。选用100只1日龄白羽种蛋鸡公雏,随机分为5组(每组20只),即空白组、CY组(环磷酰胺+生理盐水)组、高剂量组(环磷酰胺+60 mg/mL车前子黄酮)、中剂量组(环磷酰胺+40 mg/mL车前子黄酮)和低剂量组(环磷酰胺+20 mg/mL车前子黄酮),比较不同水平车前子黄酮对氧化应激鸡抗氧化能力的影响。结果显示,车前子黄酮最佳提取条件是乙醇体积分数为64.80%、料液比为1∶21.58、微波处理时间为7.08 min,该条件下黄酮的提取率为11.40 mg/g。动物试验结果表明,车前子黄酮可显著提高氧化应激鸡血清的总抗氧化能力,提高超氧化物歧化酶、谷胱甘肽过氧化物酶和过氧化氢酶的活性,并可以降低血清中丙二醛的浓度。结果表明,采用响应面法对车前子黄酮提取条件进行优化合理可行,车前子黄酮可在一定程度上提高鸡机体的抗氧化能力。
