探讨了血清谷胱甘肽S-转移酶(GST)对硫代乙酰胺(TAA)所致小鼠急性肝损伤的早期诊断意义。结果表明,腹腔注射TAA25~100mg/kg能增加小鼠血清GST活性,其活性改变与TAA之间存在量-效和时-效关系,与血清ALT变化间呈高度正相关。TAA致血清GST活性明显异常的同时,肝谷胱甘肽含量和GST、谷胱甘肽过氧化物酶和超氧化物歧化酶活性明显降低。提示肝脏抗氧化功能减退是TAA致肝损伤的主要机理之一,血清GST水平是反映TAA急性肝损伤的早期灵敏指标。
To investigate the early diagnostic significance of serum glutathione S-transferase (GST) on acute liver injury induced by thioacetamide (TAA) in mice. The results showed that intraperitoneal injection of TAA 25 ~ 100mg / kg could increase the serum GST activity in mice, and there was a dose-response and time-efficiency relationship between the activity of TAA and TAA, which was positively correlated with the change of serum ALT. TAA induced serum GST activity was significantly abnormal at the same time, liver glutathione content and GST, glutathione peroxidase and superoxide dismutase activity was significantly reduced. It is suggested that the decrease of liver anti-oxidative function is one of the main mechanisms of TAA-induced liver injury. Serum GST level is an early sensitive indicator of TAA acute liver injury.
